It's not a question of quantity with twitter, in the beginning I was hunting and reached a couple of thousand twitterers, but the results were so-so.
a.) It's better to check the followers of twitterers who are selling/marketing in a similar niche as yours, and add them.
b.) Periodically check who didn't follow you back, remove them, so you stay within the boundaries/ratio needed to continue adding users.
c.) You are going to write messages that make sense, in good plain English, and not spam people, as you will lose them otherwise.
d.) Be nice, honest and real, try to put yourself in their shoes. Nobody likes 100 messages a day with no meaning or just a link.

I would suggest you to be like a genuine user and post genuine and useful tweets.Also follow other people so that they follow you.I have seen a twitter user who was following only 2 people and he had over 300 followers.Also try to check how the others who have more followers tweet.and also use the hashtags as explained earlier and use in the hashtags the most relevant keywords.Suppose when a person searches for that particular keyword then he should find your tweets and will most probably will follow you.Also do some more research about the hashtags.I am currently having 110 followers and trying to exceed this.Also post some useful articles related to your niche.Thanks

One trick is to follow people who have the same interests as you or whatever you website is about.
